Pour ce faire, nous travaillons \u00e0 la mise en place d'une plateforme o\u00f9 les chercheurs pourront acqu\u00e9rir des comp\u00e9tences avanc\u00e9es en ing\u00e9nierie mol\u00e9culaire et contribuer au d\u00e9veloppement d'outils innovants pour contr\u00f4ler les vecteurs de maladies, tels que les moustiques responsables du paludisme.<\/p>\n\n\n\n<p>Une \u00e9tape cl\u00e9 de ce projet consiste \u00e0 renforcer l'expertise et le savoir-faire technique de notre \u00e9quipe de chercheurs au Mali. Plus important encore, elle nous aidera \u00e0 renforcer les capacit\u00e9s en ing\u00e9nierie mol\u00e9culaire sur le continent et \u00e0 nous pr\u00e9parer \u00e0 former la prochaine g\u00e9n\u00e9ration de scientifiques africains.<\/p>\n\n\n\n<p>En 2024, apr\u00e8s approbation des autorit\u00e9s r\u00e9glementaires comp\u00e9tentes, notre \u00e9quipe \u00e0 l'USTTB\/MRTC a import\u00e9 une souche de moustiques non g\u00e9n\u00e9tiquement modifi\u00e9s appel\u00e9e \"G3\" de l'universit\u00e9 de Keele. Nous commencerons par modifier cette souche en introduisant des marqueurs fluorescents (ceux-ci seront visibles dans des cellules et des tissus pr\u00e9d\u00e9finis des insectes modifi\u00e9s). Ma th\u00e8se porte sur la manipulation g\u00e9n\u00e9tique des g\u00e8nes de reproduction chez les moustiques&nbsp;<em>Anopheles gambiae,&nbsp;<\/em>l'un des principaux vecteurs du paludisme en Afrique subsaharienne. J'\u00e9tudie une approche qui pourrait \u00eatre utilis\u00e9e pour r\u00e9duire le nombre de ces moustiques et donc r\u00e9duire la transmission du paludisme. L'une des principales choses que j'ai apprises jusqu'\u00e0 pr\u00e9sent au cours de mon travail \u00e0 Keele est l'utilisation d'outils informatiques pour concevoir des constructions g\u00e9n\u00e9tiques afin d'introduire ou de modifier des g\u00e8nes chez les moustiques, comme l'insertion de marqueurs fluorescents ou le ciblage de g\u00e8nes sp\u00e9cifiques avec CRISPR\/Cas9, qui sont \u00e9galement largement applicables \u00e0 d'autres applications d'\u00e9dition de g\u00e8nes en dehors des insectes. J'ai endur\u00e9 les sympt\u00f4mes d\u00e9bilitants du paludisme et j'ai vu d'innombrables amis et membres de ma famille combattre la maladie \u00e0 plusieurs reprises. Les approches g\u00e9n\u00e9tiques de la lutte antivectorielle, telles que les moustiques g\u00e9n\u00e9tiquement modifi\u00e9s, constituent une approche prometteuse dans la lutte contre ces maladies.
